Piqray capsules What is Piqray? Piqray is a drug that is available only with a doctor's prescription and is used by men and women who have gone through menopause if they have HR-positive, HER2-negative breast cancer that has progressed or spread to other areas of the body (metastatic). Piqray is solely utilized in the event that your malignancy has a certain genetic marker (an abnormal "PIK3CA" gene). This gene will be checked for in you by your primary care physician. Piqray is administered concurrently with fulvestrant in this study (Faslodex). Warnings Piqray should not be taken if you have ever experienced a severe allergic response to alpelisib or if you have an allergy to any of the components that are contained in this drug. If you have severe diarrhea, you should see your doctor as soon as possible. Before beginning to use this medication If you have ever had an allergic reaction to alpelisib, you should not take Piqray. Tell your doctor if you have ever been diagnosed with diabetes or if you have ever suffered from a severe skin rash so that they can determine whether or not this medication is safe for you to take (skin pain, redness, peeling, burning eyes, blistering of your lips or mouth). Although though Piqray is only intended for use in women who are no longer able to become pregnant, you may be required to obtain a pregnancy test that comes back negative before beginning this therapy. Talk to your healthcare provider if you have any suspicions that you could be pregnant. If either the mother or the father is taking one of these medications while pregnant, it is possible for the unborn child to be harmed or for birth abnormalities to occur. • If you are a woman and you are pregnant, you should not use Piqray at any time. While you are taking this medication and for at least one week after you have finished your last dosage, you should avoid pregnancy by using an effective method of birth control. • If you are a man and your sexual partner is pregnant or has the potential to become pregnant, you are required to use effective birth control and condoms. Keep using birth control for at least one week after your last dose. • Inform your doctor as soon as possible if a pregnancy occurs while either the mother or the father is using Piqray. • Women who also take fulvestrant may be required to avoid becoming pregnant for at least one year after treatment has been completed. Talk to your physician about how long you should continue taking the birth control method. Both men and women may experience decreased fertility (inability to bear children) as a result of taking alpelisib with fulvestrant. However, given the potential risks posed to an unborn child by these medications, it is critical to make use of birth control methods in order to avoid becoming pregnant. Piqray should not be used if you are breastfeeding, and you should wait at least one week following your last dosage to start breastfeeding again. After receiving your final dosage of fulvestrant, you should wait at least one year before attempting to breastfeed your child. Ask your doctor. What is the recommended dosage for Piqray? Always follow your physician's instructions when using Piqray. Read all of the medication guides or instruction papers that come with your medicine, and make sure to follow all of the guidelines that are included on the prescription label. Piqray pills should be taken once day, at the same time each day, with meals. Do not crush, chew, or break the pill; it should be swallowed in its whole. Do not use a tablet that has been fractured or cracked. Piqray should not be used again if you experience nausea or vomiting soon after taking it. You won't need to take the medication again until the time of your next regularly scheduled dose. Piqray can produce severe diarrhea, which can be life-threatening if it leads to dehydration or renal damage. Piqray can also cause abdominal cramping and vomiting. If you experience diarrhea while using Piqray, you should immediately begin taking an anti-diarrhea medication such as loperamide (Imodium) in order to treat the diarrhea as rapidly as possible. Take in some additional fluids, and then give the doctor a call. Injections of fulvestrant are typically administered once every two weeks in the beginning, and then on a monthly basis after that. This injection will be administered to you by a qualified medical expert. You will need to have your blood sugar checked on a regular basis, and your physician may also want to do other blood tests on you. The chemotherapy drugs alpelisib and fulvestrant are typically used until the patient's body no longer reacts to the treatment or until the patient experiences a severe adverse effect. Your physician will decide how long your treatment should last. Keep at room temperature and away from heat and moisture when storing. Retain each pill in its individual blister pack until it is time to consume one of the tablets. What can I expect if I forget to take a dose? You should take the medication (while you are eating), as soon as you remember to, but you should omit the missed dosage if it has been more than nine hours since the scheduled time. It is not recommended to take two dosages at once. If you are going to be late for your fulvestrant injection appointment, you should call your physician to get instructions. What should be avoided Always be sure to follow your doctor's recommendations regarding any dietary, beverage, or exercise limitations. Piqray side effects You should seek immediate medical attention if you experience symptoms of an allergic reaction to Piqray, such as hives, warmth or tingling, difficulty breathing, fast heartbeats, or swelling in your face or throat. You should also seek immediate medical attention if you experience a severe skin reaction (fever, sore throat, burning eyes, skin pain, red or purple skin rash with blistering and peeling). If you experience a severe response to a medicine that can impact many different sections of your body, you should seek medical assistance. There is a possibility that you will have the following symptoms: a rash on your skin, a fever, enlarged glands, aches and pains in your muscles, extreme weakness, unusual bruises, or yellowing of your skin or eyes. You should see your physician as soon as possible if you have: • chest discomfort, coughing, and a sense of being short of breath; • severe or persistent diarrhea; • blisters or ulcers in your mouth, red or swollen gums, difficulty swallowing; • pale skin, unusual weariness, and chilly hands and feet; • little or no urine; or • any of these symptoms. • increased thirst, increased urination, dry mouth, fruity breath odor, disorientation, hunger, and weight loss due to high blood sugar; In the event that you experience certain adverse effects, it is possible that your cancer treatments will be halted temporarily or permanently. Common adverse reactions of Piqray may include the following: nausea, vomiting; lack of appetite, weight loss; feeling weak or weary; mouth sores; rash; hair loss; or abnormal blood tests. Piqray may also cause mouth sores.
